Pharmacotherapeutic group : calcium channel blocker of the dihydropyridine group.
Dosage form : tablets.
Release form : round flat tablets of white or almost white color, planimetric cellular packing, cardboard pack.
Drug that has antihypertensive, antianginal effect. It is a selective calcium channel blocker.
Active ingredient :
- amlodipine benzylate.
Auxiliary components :
- potato starch;
- microcrystalline cellulose
- magnesium stearate;
- calcium stearate;
Vero-Amlodipine prevents the entry of calcium ions through the membranes into calcium channels (vessels and smooth muscle cells of the myocardium). As a result of the expansion of blood vessels, a decrease in blood pressure occurs. It relaxes vascular smooth muscles, promotes the expansion of peripheral arterioles, reduces round focal disease, without causing the development of reflex tachycardia. Under the influence of the drug, myocardial oxygen demand and energy consumption of the heart muscle are reduced. At the same time, there is information about the ability of this drug to expand the large coronary arteries, as well as intact coronary arterioles and ischemic myocardial regions. Under the influence of the drug, platelet aggregation is inhibited, the glomerular filtration rate increases, diuresis and sodium excretion.
The effect after taking the pills develops within 1-2 hours (regardless of the meal) and lasts for a day. Providing a prolonged effect, Vero-Amlodipine promotes a smooth decrease in blood pressure. It is absorbed into the blood slowly, but almost completely, reaching a maximum concentration in 6-12 hours. To create an equilibrium concentration requires 7-8 days of continuous intake. The half-life is 35-48 hours. Able to penetrate the blood-brain barrier. Excreted in bile, urine and breast milk.
Indications for use
- Arterial hypertension (as a single agent or in combination with other drugs);
- Angina pectoris (stable, vasospastic, variable, dilated, Prinzmetala);
- Cardiomyopathy (severe forms of chronic heart failure III-IV century);
- As a monotherapy drug in the treatment of patients refractory to the effects of nitrates and β-blockers.
- Individual intolerance to the components of the drug;
- Pregnancy and lactation (relative contraindication).
Dosage and administration
Vero-Amlodipine (tablets) is a drug for internal use. With angina and arterial hypertension, the initial dose of the drug is 5 mg / day. If necessary, it can be increased to 10 mg per day.
Patients suffering from dilated cardiomyopathy, it is recommended to start taking with 2.5 mg of the drug per day, gradually increasing the dose (of course, with good endurance) to 10 mg / day. With simultaneous use of β-blockers, ACE inhibitors and thiazide diuretics, it is not necessary to adjust the dosage. The maximum daily dose of Vero-Amlodipine should not exceed 10 mg.
From the senses : dizziness, drowsiness, fatigue, headache.
On the part of the digestive tract : nausea, abdominal pain, dyspeptic symptoms, abnormal stool, jaundice.
Since the cardiovascular system : palpitations, flushing of the face, abnormal heart rhythm, chest pain, orthostatic hypotension.
On the part of the respiratory system : shortness of breath.
On the part of the urinary system: the development of peripheral edema, increased frequency of urination (rarely), impotence (very rarely).
For the skin: itching, hyperemia, urticaria , polymorphic erythema (very rare).
On the part of the musculoskeletal system : myalgia, arthralgia, paresthesia. With prolonged use of pain in the limbs.
Other disorders: gynecomastia, gingival hyperplasia, increased levels of liver enzymes.
With simultaneous use with calcium channel blockers, increased hypotensive effect is noted.
When used in combination with lithium carbonate preparations, the neurotoxicity of the latter may be enhanced.
The hypotensive effect is enhanced with simultaneous use of anesthesia.
The use of Vero-Amlodipine together with estrogens and sympathomimetics reduces the antihypertensive effect of the drug (due to sodium retention).
When interacting with amiodarone and quinidine, the hypertensive effect is enhanced.
In combination with orlistat, a decrease in the antihypertensive effect of amlodipine is observed. This can cause an increase in blood pressure and the development of hypertensive crisis.
With simultaneous use with NSAIDs, due to inhibition of prostaglandin synthesis and fluid retention, the effectiveness of the hypertensive drug is reduced.
In case of overdose of the drug, there is a long-term pronounced decrease in blood pressure, excessive peripheral vasodilation (vasodilation), tachycardia .
