Pharmacotherapeutic group : calcium channel blocker of the dihydropyridines group.
Dosage form : tablets.
Product : round flat tablets white or almost white, contour squares, cardboard tutu.
A drug that has antihypertensive, antianginal action. It is a selective blocker of calcium channels.
Active substance :
- Amlodipine benzylate.
Auxiliary components :

Vero-Amlodipine prevents the entry of calcium ions through the membranes into the calcium channels (vessels and smooth muscle cells of the myocardium). As a result of vasodilatation, blood pressure decreases. Relaxes the smooth muscles of the vessels, promotes the expansion of peripheral arterioles, reduces the OPSS, without causing the development of reflex tachycardia. Under the influence of the drug, the need for myocardium in oxygen decreases and the energy consumption of the heart muscle. At the same time, there is evidence of the ability of this drug to expand large coronary arteries, as well as intact coronary arterioles and ischemic areas of the myocardium. Under the influence of the drug, platelet aggregation is inhibited, the glomerular filtration rate, diuresis and elimination of sodium are increased.
The effect after taking the tablets develops within 1-2 hours (regardless of food intake) and lasts throughout the day. Providing a prolonged action, Vero-Amlodipine contributes to a smooth decrease in blood pressure. Absorbed into the blood slowly, but almost completely, reaching a maximum concentration after 6-12 hours. To create an equilibrium concentration, 7-8 days of continuous reception is required. The half-life is 35-48 hours. Is able to penetrate the blood-brain barrier. It is excreted with bile, urine and breast milk.
Indications for use
- Arterial hypertension (as a monotherapy or in combination with other drugs);
- Angina pectoris (stable, vasospastic, variate, dilated, Prinzmetal);
- Cardiomyopathy (severe forms of chronic heart failure III-IV century);
- As a monotherapy in the treatment of patients refractory to the effects of nitrates and β-blockers.
- Individual intolerance to the components of the drug;
- Arterial hypotension;
- Pregnancy and lactation (relative contraindication).
Dosing and Administration
Vero-Amlodipine (tablets) is a drug for internal use. With angina and hypertension, the initial dose of the drug is 5 mg / day. If necessary, it can be increased to 10 mg per day.
Patients suffering dilated cardiomyopathy, it is recommended to start taking with 2.5 mg of the drug per day, gradually increasing the dose (of course, with good tolerability) to 10 mg / day. With simultaneous use of β-blockers, ACE inhibitors and thiazide diuretics, dosage adjustment is not required. The maximum daily dose of Vero-Amlodipine should not exceed 10 mg.
From the senses : dizziness, drowsiness, fatigue, headache.
On the part of the digestive tract : nausea, abdominal pain, dyspepsia, stools, jaundice.
On the part of the cardiovascular system : palpitation, flushing of the blood to the face, violation of the heart rhythm, pain in the chest, orthostatic hypotension.
On the part of the respiratory system : dyspnea.
From the side of the urinary system: the development of peripheral edema, increased frequency of urination (rarely), impotence (very rarely).
From the skin: itching, hyperemia, urticaria , erythema multiforme (very rarely).
From the musculoskeletal system : myalgia, arthralgia, paresthesia. With prolonged use of pain in the limbs.
Other disorders: gynecomastia, gingival hyperplasia, increased level of hepatic enzymes.
With simultaneous use with calcium channel blockers, there is an increase in hypotensive effect.
When used in conjunction with lithium carbonate preparations, the neurotoxicity of the latter can be enhanced.
The hypotensive effect is intensified with simultaneous use of anesthesia.
The use of Vero-Amlodipine along with estrogens and sympathomimetics reduces the antihypertensive effect of the drug (due to sodium retention).
When interacting with amiodarone and quinidine, hypertensive action is enhanced.
In combination with orlistat there is a decrease in the antihypertensive effect of amlodipine. This can lead to an increase in blood pressure and the development of hypertensive crisis.
With simultaneous use with NSAIDs, due to the oppression of the synthesis of prostaglandins and fluid retention, the effectiveness of the hypertensive drug decreases.
When the drug is overdosed, there is a long pronounced decrease in blood pressure, excessive peripheral vasodilation (vasodilatation), tachycardia .
